ARP-7383 is a hybridoma cell line generated by the fusion of P3X63 Ag8.653 myeloma cells with immunized BALB/c mouse splenocytes that secretes an IgG1 monoclonal antibody. Recombinant HIV-1 (HXB2) Tat 86 amino acids (Tat A, Cambridge Biosciences, Inc.) was used for immunization.
ARP-7383 was raised against a synthetic peptide comprising the full length (112 amino acids)human immunodeficiency virus 2 (HIV-2) ST Vpx protein.The antibody produced by the hybridoma reacts in Western blots (a dilution of 1:200 to 1:500 is recommended), immunoprecipitation and immunofluorescence assays (the appropriate concentration must be determined by titer). The monoclonal antibody produced by ARP-7383 is available as ARP-7377.
Growth characteristics: Thaw and treat as standard cells in culture. Split at approximately 5 x 105 cells per mL (1:3). The cells grow in small clumps in solution, dependant on growth conditions. The supernatant can be harvested every 2 to 5 days. For optimal production of antibodies the cells have to be monitored.
Propagation Medium: UltraDOMA™ Protein-Free Hybridoma Medium (Lonza 12-727F) supplemented with 5% fetal calf serum
ARP-7383 is negative for Mycoplasma, bacteria and fungi. Each vial of ARP-7383 contains approximately5.3 x 106 cells per vial in UltraDOMA™ Protein-Free Hybridoma Medium (Lonza 12-727F) supplemented with 10% fetal calf serum and 10% dimethyl sulfoxide (DMSO).
